Annexin V-iFluor 594 is a probe that couples the fluorescent dye iFluor 594 with the apoptosis-indicating protein Annexin V, designed for detecting early-stage cellular apoptosis.
Featuring an excitation wavelength of approximately 590 nm and emission wavelength of 617 nm, this high-brightness fluorescent probe is compatible with various imaging techniques including confocal microscopy and flow cytometry.
Annexin V can specifically recognize and bind to phosphatidylserine (PS) that is flipped out of the cell membrane, thus enabling the labeling of early apoptotic cells.
The probe has good water solubility and low background, which is suitable for the red channel labeling in multi-color staining experiments.

【Annexin V-iFluor 594標記關于PS外翻的識別過程和原理是怎樣的？】
原理
PS通常分布于細胞膜脂質雙層的內(nèi)側，但在細胞凋亡早期，PS會從內(nèi)側翻轉到細胞膜外側。Annexin V是一種鈣離子依賴性磷脂結合蛋白，對PS具有高親和力和特異性，可與之結合。
iFluor 594是一種熒光染料，標記在Annexin V上后，可通過熒光信號（紅色）直接示蹤PS的外翻現(xiàn)象。該標記不會穿透完整細胞膜，因此僅結合于膜外暴露的PS，需與膜不透性核酸染料聯(lián)用，以區(qū)分凋亡細胞、壞死細胞及正常細胞（雙陰性）。
識別過程
樣本制備：收集待測細胞（如經(jīng)凋亡誘導處理），重懸于含Ca2?的緩沖液中（通常為1× Binding Buffer），以維持Annexin V的PS結合活性。
標記染色：向細胞懸液中加入Annexin V-iFluor 594試劑，避光孵育15–30分鐘（室溫或4°C），使探針充分結合外翻PS。
染料聯(lián)用：同步加入PI等 viability dye（如需區(qū)分壞死），避免洗滌步驟導致細胞損傷或信號丟失。
檢測分析：立即通過流式細胞術或熒光顯微鏡檢測。Annexin V-iFluor 594的紅色熒光信號（Ex/Em ≈ 590/617 nm）用于量化PS外翻水平，結合PI的綠色熒光可進一步區(qū)分細胞狀態(tài)。
